Reprogramming the tumor metastasis cascade by targeting galectin-driven networks

چکیده
A sequence of interconnected events known as the metastatic cascade promotes tumor progression by regulating cellular and molecular interactions between tumor, stromal, endothelial, immune cells both locally systemically. Recently, a new concept has emerged to better describe this process defining four attributes that should undergo. Every individual hallmark represents unique trait cell impacts directly in outcome metastasis process. These critical features, hallmarks metastasis, include motility invasion, modulation microenvironment, plasticity colonization. They are hierarchically regulated at different levels several factors, including galectins, highly conserved family β-galactoside-binding proteins abundantly expressed microenvironments sites metastasis. In review, we discuss role galectins modulating each highlighting novel therapeutic opportunities for treating disease.

Reprogramming Tumor-Associated Macrophages by Antibody Targeting Inhibits Cancer Progression and Metastasis.
Tumors are composed of multiple cell types besides the tumor cells themselves, including innate immune cells such as macrophages. Tumor-associated macrophages (TAMs) are a heterogeneous population of myeloid cells present in the tumor microenvironment (TME). Here, they contribute to immunosuppression, enabling the establishment and persistence of solid tumors as well as metastatic dissemination...
متن کاملExtracellular Galectin-3 in Tumor Progression and Metastasis
Galectin-3, the only chimera galectin found in vertebrates, is one of the best-studied galectins. It is expressed in several cell types and is involved in a broad range of physiological and pathological processes, such as cell adhesion, cell activation and chemoattraction, cell cycle, apoptosis, and cell growth and differentiation. However, this molecule raises special interest due to its role ...
متن کاملTargeting tumor metastasis by regulating Nm23 gene expression.
The Nm23 gene is a metastatic suppressor identified in a melanoma cell line and expressed in different tumors where their levels of expression are associated with reduced or increased metastatic potential. Nm23 is one of the over 20 metastasis suppressor genes (MSGs) confirmed in vivo. It is highly conserved from yeast to human, implying a critical developmental function. Tumors with alteration...
متن کاملGalectin-9 suppresses tumor metastasis by blocking adhesion to endothelium and extracellular matrices.
We previously described an inverse correlation between galectin-9 (Gal-9) expression and metastasis in patients with malignant melanoma and breast cancer. This study verified the ability of Gal-9 to inhibit lung metastasis in experimental mouse models using highly metastatic B16F10 melanoma and Colon26 colon cancer cells. B16F10 cells transfected with a secreted form of Gal-9 lost their metasta...

A 50 years old woman with history of pancreatic neuroendocrine tumor diagnosed 2 years ago, which has not been surgically removed, was referred to our department for a 99mTc-Octreotate in order to evaluate the somatostatin receptor status. She was treated with regular sandostatin injections and chemotherapy. Her CT scan which was previously performed confirmed lung, adrenal and hepati...
